HEPATOPROTEKTIF EKSTRAK KULIT DELIMA (Punica Granatum L) PADA TIKUS YANG DIINDUKSI CCL4

Abstract

Penelitian dilakukan untuk mengetahui dosis efektif ekstrak kulit delima terhadap kerusakan hati dengan indikator kadar Serum Glutamik Piruvat Transaminase (SGPT) dan malondialdehid (MDA) plasma pada tikus jantan yang diinduksi karbon tetraklorida (CCl4). Target penelitian ini adalah dapat dibuktikannya efek penghambatan ekstrak kulit delima terhadap kerusakan hati tikus akibat induksi CCl4 serta mendapatkan dosis optimal. Penelitian bersifat laboratorik dengan rancangan acak lengkap, menggunakan sampel 36 ekor tikus dengan jumlah replikasi setiap kelompok adalah 4 ekor. Objek dikelompokkan dalam 9 kelompok yang akan membandingkan efek ekstrak kulit delima mulai dari 100mg/KgBB hingga 600mg/KgBB dosis tunggal yang diberikan 3 jam sebelum tikus diinduksi CCL4 peroral . Indikator penghambatan kerusakan hati ditinjau dari kadar SGPT dan MDA plasma. Analisis data menggunakan uji Kruskal Wallis dilanjutkan uji Mann-Whitney. Hasil penelitian menunjukkan kelompok perlakuan dengan ekstrak kulit dlima dosis 300 mg/kgBB hingga 600 mg/kgBB memiliki kadar SGPT dan MDA plasma lebih rendah secara bermakna (P<0,05) dibandingkan dengan kontrol positif yang diberi CCl4. Ekstrak kulit delima dosis 500 mg/kgBB paling efektif menghambat peningkatan kadar SGPT, sedangkan dosis 400 mg/kgBB dan 500 mg/kgBB mampu menghambat peningkatan kadar MDA plasma dengan efektivitas yang sama (p>0,05). Ekstrak etanol kulit delima menghambat peningkatan kadar SGPT dengan dosis paling efektif 500 mg/kgBB dan menghambat kadar MDA plasma dengan dosis paling efektif 400 mg/kgBB.
